When using a Big Muff, I find myself cranking the Sustain knob and parking it there. If I wanted to make a Big Muff with sustain permanently maxed out, could I just remove R6, R7, and C4 (and connect C3 to R8)? Based on this schematic: http://www.generalguitargadgets.com/pdf/ggg_bmp_vram_sc.pdf

the q2 stage shows a 100nF//8k2 connection to the q3 stage, so your mod looks about right.
If you want the circuit to be exactly like the stock circuit with the sustain maxed, I would just remove the sustain control, and connect the 1K to ground between the caps.

That would be needed to emulate the sustain control in it's lowest setting....and we don't want that......

Make it a 100k to ground and what Larry said makes sense, that would simulate what ever effect the pot has when turned up full. Probably not needed but there may be some difference in sound between R6 being 101k Ohms and being infinite Ohms.
Another thing which might matter is C3 and C4 in series give a total capacitance of 0.05uF, so if you remove C3 you might want to make C4 0.047uF to compensate. Again might make no odds.
